What is it?

OH! It was an old mod for Warcraft 3 that we used to play back in the day after EQ raids. It was called Defense of the Ancients (DotA). It became very popular, and I honestly had more fun on it then WC3 at times. The problem was it was always just a mod. Valve finally took it over and made an actual game of it that allows the fans to submit customized item graphics and such.

It's a simple game where each side has a "base" that must be destroyed. There are 3 main lanes leading between each base. Every 30 seconds, a few neutral critters will spawn in each lane for each side and move down the lanes and fight. If things stay equal neither side makes headway. There are towers that do a lot of damage on each sides half that must be dealt with to get into the base. Basically, each team has 5 players. There are over a 100 heroes you can play with, and hundreds of items and item combinations. You acquire gold by killing critters and enemy heroes, as well as experience and level up and buy new and better items, etc - each side tries to slowly gain momentum and push into and destroy the other base. It's simple, but incredibly fun. Strategies for the game have become very complex at the pro level.
